چکیده
ABSTRACT Problem Multiple etiologies are available for understanding the mechanisms responsible for the syndrome of Preeclampsia. Cytokine imbalance and endothelial dysfunction is suggested to have a pivotal role. The pathophysiological processes in preeclampsia (PE) are also influenced by genetic factors. To characterize the host genetic factors determining the susceptibility to Preeclampsia, we evaluated the influence of polymorphism in eNOS gene on nitric oxide and cytokines production and on the risk of developing PE. Experimental approach This cross-sectional study included 150 women with preeclampsia and 150 healthy pregnant women. Their blood samples were analyzed for nitric oxide, inflammatory cytokines and eNOS gene polymorphism. Major findings Decreased NO (reflecting endothelial dysfunction) and increased cytokines (interleukin-6, Tumor Necrosis Factor-α, interleukin-2 and Interferon-γ) levels were found in preeclampsia (p<0.001). Significant differences were found in genotype/allele distribution between two groups. Significant negative correlation was seen between NO and cytokines levels (IL-6, TNFα, INFγ & IL-2) in preeclamptic group (p=0.001). Conclusion We concluded that endothelial dysfunction and inflammatory markers could be involved in the pathogenesis of Preeclampsia and those markers could be correlated with genotypic variations.

Association of Endothelial Dysfunction with Endothelin, Nitric Oxide and eNOS Glu298Asp Gene Polymorphism in Coronary Artery Disease
The endothelial dysfunction has been implicated as a major event in the pathogenesis of atherosclerosis. Therefore, this study was planned to determine (a) role of endothelium-derived nitric oxide (NO) and endothelin as coronary artery disease (CAD) risk markers and (b) intergenotypic variation of endothelial nitric oxide synthase (eNOS) Glu298Asp polymorphism in CAD.The endothelin, NO and eNOS...
متن کاملAssociation of endothelial nitric oxide synthase gene G894T polymorphism with type two diabetes and diabetic nephropathy
Background: Nitric oxide (NO) produced by endothelial NO synthase (eNOS) mediates a large range of processes, and abnormality in the production of NO has been implicated in diabetic complications including diabetic nephropathy (DN). G894T polymorphism in the eNOS gene has been shown to decreased activity the NO levels of plasma. The association between eNOS Glu298Asp gene polymorphism and DN ri...

Background Varicocele is an abnormal dilation and tortuosity of veins of pampiniform plexus that drains the testis and causes an important change in semen. This abnormality is often one of the most common risk factors for male infertility. The aim of this study was to investigate the relationship between nitric oxide synthase 3 (NOS3) 4a4b (rs61722009) gene polymorphism, as a common genetic fact...
